He became a registered practitioner in Queensland on December 24, 1891. An advertisement in the Brisbane Courier during January 1892 stated that he was a “homœopathic physician”, and could be consulted at No. 4 Hodgson Terrace, George Street. In the post office directory of  1894 he was listed as being at Wickham Terrace.

By 1895 he was located at Georgetown, and during August of that year was married to Miss Maude Sheridan at Petersham in Sydney. In Georgetown he became a Commissioner of the Peace and was surgeon at the 12 bed Etheridge District Hospital. In 1899 and 1911 he was listed as residing and practising in Cairns.

Knowles Street in Babinda is named after Dr Knowles, who at one time owned large areas of the district. Knowles Street in Whitfield is also named after him.
